Evaluation of portable XRF instrumentation for assessing potential environmental exposure to toxic elements

Abstract
Portable instruments based on X-Ray Fluorescence Spectrometry (XRF) have the potential to assist in field-based studies, provided that the data produced are reliable. In this study, we evaluate the performance of two different types of XRF instrument (XOS prototype and Thermo Niton XL3t). These two XRF analysers were evaluated in a laboratory setting, and data were reported for 17 elements (As, Ba, Cd, Co, Cr, Cu, Fe, Hg, Mn, Ni, Pb, Se, Sn, Sr, Ti, V, and Zn). Samples analysed (n = 38) included ethnic herbal medicine products (HMPs), ethnic spices (ES), and cosmetic products (CPs). Comparison analyses were carried out using Inductively Coupled Plasma Optical Emission Spectrometry (ICP-OES). In general, results reported for Cd, Cu, and Pb by the XOS prototype analyser, using the non-metal mode, were negatively biased (5–95%) as compared to ICP-OES. In contrast, results reported for Pb, As, Cd, Cu and Zn by the Niton, using the soil mode, were positively biased, in some instances (Cd) by up to four orders of magnitude. While the sensitivity of both instruments was insufficient for reliably ‘quantifying’ toxic elements below 15 mg/kg, XRF was still capable of positively ‘detecting’ many elements at the low single-digit mg/kg levels. For semi-quantification estimates of contaminants at higher levels, and with limited sample preparation, both XRF instruments were deemed fit for the purpose. This study demonstrates that modern XRF instrumentation is valuable for characterising the elemental content of food, cosmetic, and medicinal products. The technology is particularly useful for rapidly screening large numbers of products (100’s per day) in the field, and quickly identifying those that may contain potentially hazardous levels of toxic elements. Toxic elements can be confirmed by examining the raw spectrum, and the limitations of factory-based calibration are generally manageable for field-based studies. © 2015 Wadsworth Center, New York State Department of Health.

Immune response and innervation signatures in aseptic hip implant loosening

Abstract
Background: Aseptic loosening (AL) of hip prosthesis presents inflammation and pain as sign and symptom similarly to arthritis pathologies. Still, the immune and innervation profiles in hip AL remain unclear and their interplay is poorly explored. Herein, local tissue inflammatory response, sensory and sympathetic innervation as well as associated local mediators were assessed in hip joint microenvironment underlying AL and compared to osteoarthritis (OA). Methods: Histopathological analysis, immune cells (macrophages, T, B cells and PMNs) as well as sensory and sympathetic nerve fibers (SP+, CGRP(+), TH+) distribution and profiles were analyzed on tissues retrieved from patients with failed hip prostheses due to AL (n = 20) and hip OA (n = 15) by immunohistochemistry. Additionally, transcriptional levels of pro-inflammatory cytokines (TNF-alpha, IL-1 beta, IL-6, IL-12a, iNOS), anti-inflammatory cytokine (IL-10), osteoclastic factor (RANKL) and bone remodeling factor (TGF-beta 1) were locally evaluated by qRT-PCR. Serum TGF-beta 1 levels were assessed preoperatively by ELISA. Results: Histopathological analysis revealed that tissues, aseptic interface membranes of AL patients had distinct tissue architecture and immune cells profile when compared to OA synovial tissues. Macrophages, T cells and B cells showed significant differences in tissue distribution. In OA, inflammation is mostly confined to the vicinity of synovial membrane while in AL macrophages infiltrated throughout the tissue. This differential immune profile is also accompanied with a distinct pattern of sensory and sympathetic innervation. Importantly, in AL patients, a lack of sympathetic innervation aseptic interface membranes without compensation mechanisms at cellular levels was observed with simultaneous reorganization of sensorial innervation. Despite the different histopathological portrait, AL and OA patients exhibited similar transcriptional levels of genes encoding key proteins in local immune response. Nevertheless, in both pathologies, TGF-beta 1 expression was prominent in sites where the inflammation is occurring. However, at systemic level no differences were found. Conclusion: These findings indicate that AL patients exhibit different local inflammatory response and innervation signatures from OA patients in hip joint. These insights shed the light on neuro-immune interplay in AL and highlight the need to better understand this crosstalk to unravel potential mechanisms for targeted-therapies to improve hip joint lifetime and treatment.

A surveyonheuristics for the two-dimensional rectangular strip packing problem

Abstract
Two-dimensional rectangular strip packing problems belong to the broader class of Cutting and Packing (C&P) problems, in which small items are required to be cut from or packed on a larger object, so that the waste (unused regions of the large object) is minimized. C&P problems differ from other combinatorial optimization problems by the intrinsic geometric constraints: items may not overlap and have to be fully contained in the large object. This survey approaches the specific C&P problem in which all items are rectangles, therefore fully characterized by a width and a height, and the large object is a strip, i.e. a rectangle with a fixed width but an infinite height, being the problem’s goal to place all rectangles on the strip so that the height is minimized. These problems have been intensively and extensively tackled in the literature and this paper will focus on heuristic resolution methods. Both the seminal and the most recent approaches (from the last decade) will be reviewed, in a rather tutorial flavor, and classified according to their type: constructive heuristics, improvement heuristics with search over sequences and improvement heuristics with search over layouts. Building on this review, research gaps are identified and the most interesting research directions pointed out. © 2016 Brazilian Operations Research Society.

Abstract
Machine learning has been well applied and recognized as an effective tool to handle a wide range of real situations, including medical applications. In this scenario, it can help to alleviate problems typically suffered by researchers in this field, such as saving time for practitioners and providing unbiased results. This tutorial is concerned with the use of machine learning techniques to solve different medical problems. We provide a survey of recent methods developed or applied to this context, together with a review of novel contributions to the ESANN 2016 special session on Machine learning for medical applications.

Abstract
Background: Malnutrition is a modifiable risk factor for several diseases among elderly. To develop appropriate nutrition policies to decrease malnutrition, up-to-date information on the dimension of the problem is warranted. Objective: To estimate the prevalence of malnutrition among Portuguese elderly (65+ years old) resident in nursing homes.

An overview of project data for integrated project management and control

Abstract
In this paper, an overview is given of the project data instances available in the literature to carry out academic research in the field of integrated project management and control. This research field aims at integrating static planning methods and risk analyses with dynamic project control methodologies using the state-of-the-art knowledge from literature and the best practices from the professional project management discipline. Various subtopics of this challenging discipline have been investigated from different angles, each time using project data available in literature, obtained from project data generators or based on a sample of empirical case studies. This paper gives an overall overview of the wide variety of project data that are available and are used in various research publications. It will be shown how the combination of artificial data and empirical data leads to improved knowledge on and deeper insights into the structure and characteristics of projects useful for academic research and professional use. While the artificial data can be best used to test novel ideas under a strict design in a controlled academic environment, empirical data can serve as the necessary validation step to translate the academic research results into practical ideas, aiming at narrowing the bridge between the theoretical knowledge and practical relevance.
